PBS buffer solution is one of the most widely used buffer solutions in biochemical research. Its main components are Na₂HPO₄, KH₂PO₄, NaCl and KCl. Generally, it serves as a solvent and plays the role of dissolving and protecting reagents. Since Na₂HPO₄ and KH₂PO₄ have secondary dissociation, the buffered pH value range is very wide, and the main function of NaCl and KCl is to increase the concentration of salt ions. If necessary, 1 mmol/L CaCl₂ and 0.5 mmol/L MgCl₂ can also be added to PBS to provide divalent cations.
Function
PBS is the abbreviation of phosphate buffered saline. Not only pseudorabies virus needs to be diluted with it, but also general active biological agents need to be diluted with it. The reason is that it has a salt balance and an adjustable and suitable pH buffering effect. Distilled water does not have a salt balance effect and can damage the structure and biological properties of biological proteins. Normal saline does not have the function of adjusting the pH and cannot ensure that complete and active substances participate in biological reactions under the most suitable conditions. Therefore, using PBS is the first choice.
