The E. coli expression system is the preferred host for expressing proteins with small molecular weights or relatively simple structures, primarily used to produce cytokine-based drugs such as insulin, interferons, and interleukins. The content of E. coli residual DNA in these drugs must be strictly controlled. Additionally, in the fields of cell and gene therapy, E. coli is used in fermentation tank technology during production to amplify viral plasmids. The amplified plasmids must undergo quality control for E. coli residual DNA before being used to infect 293 cells for viral packaging. As a host cell for various biological products, nucleic acid residues from E. coli cannot be ignored.
